为了进一步探讨人精于mr在精卵融合中的作用,本文采用改良后的甘露糖-琼脂糖凝胶亲和层析法分离纯化人精子mr ,并将提纯的人精子甘露糖受体( purifiedmannosereceptor , pmr )作用于去透明带的金黄地鼠卵母细胞,运用透射电子显微镜技术和罗丹明偶联的兵豆凝集素( tritc - lca )免疫荧光标记技术观察pmr对卵子的影响。 - The results were as follows : ( l ) the pre - incubation of pmr induced the exocytosis of the cortical granules ( cgs ) in the oocytes and reduced the quantity of the microvillus on the surface of oocytes ; ( 2 ) the matrix of cg might migrate to the surface of plasma membrane of oocytes after pmr pre - treatment . ( 3 ) incubation of zona - free hamster eggs with spermatozoa pretreated with pmr a ntiserum was associated with a dose - dependent significant reduction in the number of spermatozoa binding to the oolemma , compared with a contraband with a dose - dependent reduction of the fertilization rate and the penetration index . the results indicate that mr of human sperm plays an important role in sperm - oocyte fusion and oocyte activation
结果显示: ( 1 )人精子mr能够诱发去透明带金黄地鼠卵母细胞中皮质颗粒的胞吐,并引起卵母细胞膜表面微绒毛形态和数量发生改变; ( 2 )皮质颗粒胞吐后,其内容物可结合到卵膜表面; ( 3 )抗mr抗血清预处理精子可抑制精子与卵子的结合与融合,使受精率、穿透指数和结合指数下降,并均有剂量依赖性。以上结果提示,人精子mr在受精过程中起重要作用,其主要作用可能是介导精-卵膜融合并促进卵母细胞的活化。
